Conditions: Diabete Mellitus, MASLD
Sex: All
Ages: 21 Years – 70 Years
Healthy volunteers: Yes
Enrollment: 270
Sponsor: National University Health System, Singapore

Summary
The RESET-INSPIRE study is designed to investigate individual variability in glucose response and insulin sensitivity, a key mechanism underlying metabolic-associated steatotic liver disease (MASLD) and cardiometabolic disorders. Building on findings from Project RESET, this study aims to develop and validate a continuous glucose monitoring (CGM)-based approach for assessing insulin sensitivity.
Eligibility Criteria
Inclusion Criteria:
1. Adults aged between 21 and 70 years (including 70 yrs old) at the time of screening
2. Able to speak and understand English
3. No past history of acute myocardial infarction (heart attack), stroke or heart failure
4. Able to understand and voluntarily provide written informed consent prior to any study specific procedures.
5. No current diagnosis of type 1 or type 2 diabetes, and not taking glucose-lowering medications.
6. Willing and able to comply with all scheduled study procedures.
Exclusion Criteria:
1. Known diagnosis of diabetes mellitus (type 1 or type 2), or use of antidiabetic medication.
2. Current or recent (within 3 months) use of medications known to affect glucose, including but not limited to corticosteroids.
3. Known history of liver disease or abnormal liver function deemed clinically significant by the investigator who is a medical doctor.
4. Renal impairment, defined as eGFR ≤ 60mL/min/1.73m\^2
5. Known pregnancy, plans to become pregnant during the study period, or currently breastfeeding, or
6. Known allergy or intolerance to CGM adhesives or inability to comply with CGM sensor placement, or
7. Any medical, psychiatric, or social condition that, in the opinion of the investigator, would interfere with the participant's ability to adhere to study procedures or would pose additional risk.
